Decaffeinated coffee and green tea extract inhibit foam cell atherosclerosis by lowering inflammation and improving cholesterol influx/efflux balance through upregulation of PPARγ and miR-155

Background:Foam cells are markers of atherosclerosis and characterise advanced atherosclerotic plaque, stimulated by inflammation caused high lipid levels in macrophages. The combination decaffeinated coffee green tea extract (DCGTE) has been suggested to have a role foam cell inhibition.Objective: we investigated the inhibiting DCGTE against formation, through modulation process cholesterol metabolism macrophage colony stimulating factor- (M-CSF) oxidized low-density lipoprotein (oxLDL)-exposed macrophages.Methods: Coffee were extracted filtration infusion respectively, underwent decaffeination using active carbon blanching methods, respectively. Cells administered 160/160 320/320μg/ml DCGTE. Foam formation was observed light microscope after staining with Oil Red O (ORO), accumulation lipids macrophages ELISA. Observations influx efflux determined semiquantitative cluster differentiation 36 (CD36) ATP binding cassette transporter A1 (ABCA1) expression immunofluorescence. quantified inflammatory/anti-inflammatory markers, e.g., tumor necrosis factor α (TNFα) interleukin 10 (IL10) Peroxisome proliferator activated response γ (PPARγ) activity assessed PCR ELISA, microRNA 155 (miR-155) examined qPCR.Results:DCGTE at above concentrations tended reduce numbers, significantly inhibited (p=0.000), reduced CD36 (p=0.000) TNFα secretion Raw264.7 exposed M-CSF 50ng/ml oxLDL 50μg/ml. PPARγ (p=0.00) (p=0.001), miR-155 relative IL10 production also increased.Conclusion:DCGTE lowered possibly attenuation inflammatory improvement lipid/efflux mechanisms oxLDL-stimulated cells, via upregulation miR-155. Our results suggest may help prevent atherosclerosis-based diseases.
